Our functional immortalization technology – CI-SCREEN has been recently described in Nature Communications.


CI-SCREEN is universal. It can be applied to any cell type from any mammalian species. So far we have established novel cell lines from more than 20 cell types and from 8 different species.



Importantly, CI-SCREEN not only immortalizes primary cells. CI-SCREEN keeps them functional.



Functionality has been demonstrated by several papers in peer-reviewed journals like alveolar epithelial cells (Kuehn et al., 2016), endothelial cells (Heiss et al., 2015), osteoblasts (Pérez-Campo et al., 2017), intestinal epithelial cells (Schwerk et al., 2013) and mouse embryo fibroblasts (Stichling et al., 2018).


How we work


We are flexible! You can either provide us primary cells of interest or we will acquire them for you. Primary cells are not available? 1st Step Immortalization of primary material

We will transduce the desired primary cells with our CI-SCREEN gene library. After transduction the cells are simply expanded. Thereby, we automatically screen for those cells that received an advantageous gene combination and become immortalized. Robustly proliferating cell lines are generated within 60-90 days using this process. The novel cell lines that are established harbor different gene combinations and thus all display different phenotypes.

2nd Step Characterization of the novel immortalized CI-SCREEN cell lines

The novel cell lines that are developed in the first step all harbor different gene combinations. Thus, they all display different phenotypes. Therefore the second step is to identify the desired cell line with the desired functionality. Together with the customer a cell type specific characterization scheme is worked out. Thereby the immortalized cell line(s) that match the desired phenotypes are cherry-picked.


Cell type specific markers can be e.g. analyzed by flow cytometry. The endothelial cell line (CI-huVEC) was comprehensively characterized and compared side-by-side with the primary huVEC they were derived of (CD31, Tie1, Tie2, VEGFR2, ICAM, CD144, E-Selectin, Endoglin). For details please see the Nature Communications paper (Lipps et al., 2018).


We also successfully used immunocytochemistry to analyze cell type specific markers. The alveolar epithelial cell line (CI-hAELVi) displays nice membrane bound expression of Zo-1 and E-cadherin (Kuehn et al., 2016) or as another example the chondrocytes (CI-huChon) cell line expresses Sox9 as well as Collagen II (Lipps et al., 2018). Of course, also other technologies like ELISA, Western Blotting or qRT-PCR can be done.

Functionality is important! That is why we analyze it. We test the CI-SCREEN immortalized cell lines with functional assays that are cell type specific. No matter if it is barrier formation (alveolar epithelial cell line (CI-hAELVi), lung epithelial cells (CI-huAEC), intestinal epithelial cells (CI-muINTEPI); Schwerk et al., 2013), tube formation, cytokine induced proliferation (endothelial cells (CI-huVEC); Lipps et al., 2018), Ca2+ deposits, alkaline phosphatase activity (osteoblasts (CI-huOB); Pérez-Campo et al., 2017).
